The engineers can make you what you want to whatever depth you want.
The big difference between glass/saphire and metal is that in failure metal will bend, at some point affecting the working of the watch, and glass will fracture, you can imagine. But both can be used just as well considering the small surface of a watch. There is an even bigger surface of glass on the dial side that has to withstand the pressure in any case. The glass back will be somewhat thicker 'though.
It is nothing more than a question if you want your customers to be able to see the movement or not.
Rolex doesn't want you looking.

To be objective, the weak points in the construction of a waterproof watch are the gaskets. Adding a display back does add one gasket to the 4 that are already present in a standard case(crystal/case-frame, case-frame/case-back, case-frame/tube, tube/crow...
